Both fighters are undefeated in 11 contests but McDonnell - twin brother of former world champion Jamie - will be favourite after winning a final eliminator against Josh Wale in September.
However, Wood - naturally the bigger man - had a busy year in 2013, winning on five occasions picking up British and International Masters titles in his native Nottingham.
"It's a fantastic 50-50 domestic scrap," said promoter Eddie Hearn. "Both lads are very popular and we're expecting big support from both fighters and the winner can expect an exciting reign as champion in a fantastic division."
The fight takes place on a superb night of action at Hull's Ice Arena, heading by the return of Luke Campbell in his fifth professional outing.
Darren Hamilton will defend his British light-welterweight strap against Curtis Woodhouse, while local hope Tommy Coyle defends his IBF international belt against a yet-to-be named opponent.
Veteran super-middleweight Tony Dodson will also be in action alongside local talents talents Zak Collins, Joel Haigh and Charlie Paynton.
