Collazo, a former holder of the WBA strap, had fallen out of the world picture after his loss to Andre Berto for the WBC title in 2009.
The 32-year-old went on to be stopped by Freddy Hernandez in 2011, however he has now strung together a run of four wins and holds the WBA international belt.
However the manner of his win over Ortiz, a former world champion himself after he beat Berto for the WBC belt, could now find himself back among the best in the division.
Ortiz has now lost his last three fights since beating Berto, having been knocked out in four rounds by Floyd Mayweather Jnr while a broken jaw forced him to retire against Josesito Lopez.
Ortiz (29-5-2) had hoped to kick-start his own career against Collazo - and he started brightly and looked to impose himself on the New Yorker.
However Collazo (35-5) was happy to box off the backfoot and it was a beautiful counter right as Ortiz came in that ended the fight with a second left of the second round.
It was the 18th stoppage of Collazo's career, with "The People's Champ" now setting his sights on a possible showdown with Mayweather later this year.
