Johnson, who lost on points against Fury in 2012, has been called in as a late replacement to face Chisora at the Copper Box Arena in London.
Chisora is another man chasing a return bout with Fury, having also lost a verdict against him, and is expected to battle his British rival again in the summer.
But the Atlanta man is determined to ruin these plans and expects to earn another crack at Fury for himself, if he claims an upset win over Chisora.
He told worldboxingnews.net: "A win over Chisora puts me back in there and hopefully I can get another fight with Tyson instead.
"I'm looking to speak to Peter Fury and Tyson about replacing Chisora in the summer once I take him out on Saturday night.
"Since I was only given eight days notice the first time, I think I deserve another chance. I just need to prove it against Chisora first."
Johnson is on friendly terms with Fury after they shared the ring and will ask his former foe for advice on how to defeat Chisora.
"Chisora has been involved in some very good fights," said Johnson. "He had a very good stoppage against Malik Scott and he is digging deep to try and become a champion before I do.
"I have to stop him from doing that and will be talking to Tyson about Chisora before we fight.
"Tyson is not my enemy right now as that is only when we fight. We have trained together in the past and he is a cool dude outside of the ring."
