The 31-year-old becomes the fourth player in a week to pledge his future to Derbyshire, following their surprise promotion to Division One.
Ross Whiteley, Tony Palladino and Jonathan Clare also signed three-year deals after their stunning title-winning season.
Durston is the most experienced player on the staff at the County Ground having made his debut for Somerset back in 2002.
He signed for Derbyshire from the Unicorns midway through the 2010 season and has since become a lynchpin of the team in all three competitions.
He scored 1,367 runs in all competitions last season as well as snaring 33 wickets with his occasional off-spin.
Durston said: "I am over the moon to be committing the next three years of my career to Derbyshire. This is as tight-knit a squad as I've known throughout my career and I'm delighted to be remaining a part of it for the foreseeable future.
"We have a young and extremely talented group of players and I hope my experience can help the younger guys - and the squad as a whole - to achieve more in the years ahead."
