Holder, who was in their Champions Trophy squad but did not play, replaces Ravi Rampaul who sprained his ankle in the one-wicket win over Sri Lanka.
Rampaul had a scan after that game, missing the following win over India, and the results have shown that he needs to rest the injury.
Holder debuted for the West Indies against Australia earlier this year and also impressed in limited opportunities for the Chennai Super Kings in the IPL.
Captain Dwayne Bravo had also missed that game, due to a groin strain, but his injury was not severe and he is likely to be fit for the remainder of the series.
Updated squad: Dwayne Bravo (capt), Darren Bravo, Chris Gayle, Johnson Charles, Kieron Pollard, Marlon Samuels, Denesh Ramdin, Devon Smith, Darren Sammy, Sunil Narine, Tino Best, Jason Holder, Kemar Roach
