Clarke, 31, was a key player in last season's title-winning campaign, playing in every match and scoring 760 runs at 44.70 and chipping in with 20 wickets.
His outstanding form was rewarded with a place on the England Lions tour of Australia over the winter.
The Bears' Director of Cricket, Dougie Brown, said: "Rikki was instrumental in last year's LV= County Championship success.
"He's arguably playing the best cricket of his career so it's no wonder that he's now firmly back in the minds of the England selectors.
"Having a player in your side that consistently scores big runs at number seven and bowls in excess 85mph is a massive asset to any team, and all players and staff at Edgbaston are delighted that he's committed his long-term future to Warwickshire."
Clarke said: "I've talked quite a bit over the last few months about how much I love playing for Warwickshire and how Jim Troughton, Graeme Welch, Dougie Brown, and previously Ashley Giles have made a massive impact on my career.
"However, I wanted to take my time over signing this extension as it's come at a very important time in my career and I needed to ensure that it's right for my family.
"I'm thrilled that I've been able to secure a new deal. We've got a great set-up and coaching team here at Edgbaston, many members of the squad have signed long-term contracts, and I expect us to be able to challenge for more trophies in the future."
