The Yorkshire batsman made 166 at the top of the order to help the EPP, replying to DY Patil Academy's first-innings total of 204, reach 484-6 by the close of day two.
Root hit 22 fours and a six to show he is in good form should he need to be called upon by the full England team during the current series against India.
He shared in a stand of 104 with fellow Test squad member Eoin Morgan (47) before combining with Nottinghamshire's James Taylor (72) to put on 135.
Essex youngster Ben Foakes ended proceedings unbeaten on 90, combining with Durham leg-spinner Scott Borthwick (35 not out) in an unbroken alliance worth 116 that put the EPP side into a strong position.
Steven Finn went some way to proving his match fitness following a thigh strain by taking four wickets on the opening day of the contest.
The Middlesex paceman has missed the first two matches of the series with a thigh strain but is hoping to press his claims for a recall ahead of next week's Test at Eden Gardens in Kolkata.
