Bury defeated Walsall on penalties in the first round of the tournament after a 0-0 draw at the Banks's Stadium, but have a number of injury worries for Tuesday's tie.
Strikers Andy Bishop (groin), Andy Morrell (ankle) and Danny Carlton (achilles) all miss out, while Paul Scott (thigh) and Damien Allen (groin) are also sidelined.
Manager Alan Knill will fancy his chances of causing an upset, with Tranmere having lost eight of their opening 11 league games.
