Barcelona midfielder Fabregas has a knee injury and will be replaced in Vicente del Bosque's squad by club team-mate Marc Bartra.
The absence of Costa is the interesting one, though, as the Brazilian was due to make his first appearance in a Spain shirt after opting to play for them, despite making previous friendly appearances for the country of his birth.
A statement on the Spain team's official website said Costa had a thigh injury and had been replaced by Juventus striker Fernando Llorente.
Costa must now wait to make his Spain debut, with Monday's events only adding to the saga surrounding him after his surprise decision to turn his back on the Selecao.
La Liga leaders Barcelona confirmed on their official website on Monday that the 26-year-old Fabregas would be sidelined for seven days after spraining ligaments in his right knee during Sunday's 4-1 victory over Real Betis - in which he scored a brace.
