The Seville side finished fourth in La Liga after an impressive season, and have now inked a five-year deal with the Uruguayan international after qualifying for the continent's premier competition.
The left-sided forward had been a popular figure at Estadio El Sardinero ever since his move from Nacional in 2000, and scored 23 goals over the past four seasons.
Manchester United were understood to be tracking the 26-year-old's progress, although after speedy negotiations Betis have won the race for his signature and will officially present him next week.
Regueiro's departure paves the way for Racing to move for fellow Uruguayan Dario Silva.
The 32-year-old has failed to hold down a place with Sevilla since moving from fellow La Liga outfit Malaga, and, after scoring just twice this season, is expected to depart.
