McShane has headed to The Withdean Stadium on a deal until the end of December in search of first-team experience.
The 19-year-old has yet to make his debut for United and enjoyed a spell on loan at Walsall last season.
Swindon Town were keen to sign the centre half earlier this summer but have lost out to Brighton for his signature.
Seagulls boss Mark McGhee admits his capture of McShane ends his search for a new defender.
"We're pleased to have Paul joining us," McGhee told the club's official website. "He's exactly the type of player we want at this time and we think he'll be good for us.
"We've had him watched, and he played well, so we're looking forward to working with him."
McGhee is also running the rule over French midfielder Sebastien Carole, who has already had a trial at Southampton in pre-season.
A one-month loan deal for Wayne Henderson is close to being finalised, with the Aston Villa goalkeeper possibly making his debut, along with McShane, against Derby County this weekend.
Brighton are also still interested in Southampton shot-stopper Alan Blayney but McGhee concedes a deal is unlikely to be struck this week.
