Argentine stopper Samuel appears to be high on Chelsea's wanted list and has declared his willingness to move to Stamford Bridge.
Roma's uncertain financial situation has increased the likelihood that they will have to offload Samuel in the summer.
The Blues are first in the queue and have now registered their interest with Roma, although they are yet to lodge a firm offer for the centre half.
"I can confirm that Chelsea are interested in Walter Samuel," Roma's sporting director Franco Baldini told PA International.
"We have been in contact with the English club and they have requested information on the player but they have made no official offer yet."
While Samuel appears to be heading towards the Stadio Olimpico exit door, Roma are determined to retain the services of key forward Totti.
Spanish speculation claimed Real Madrid had reached agreement on a four-year contract with the Italian international.
But despite the resurrection of Real's interest in Totti, Baldini remains unperturbed by the latest bout of rumour-mongering.
He added: "We are tranquil. These rumours about Totti don't trouble us because they are absolutely groundless."
