The Principality outfit have a 3-1 lead going into the second leg at Stamford Bridge on Wednesday.
The main doubt for the clash is wing star Rothen, who has a groin problem.
Rothen limped out of Monaco's 2-1 win at Nice on Friday after suffering a groin injury.
It remains a race against time as to whether Rothen will now be fit and coach Didier Deschamps is likely to give the midfielder as much time as possible to be fit.
Squillaci looks set to be fit for Chelsea after being rested for the game in Nice.
The centre back was suffering from a recurrence of a thigh problem, but the rest looks to have done the job for the defender.
Prso, who like Rothen was forced off early at Nice, is set to be play through the pain barrier to make the match in West London.
The Croat (pictured) broke a toe, after he opened the scoring - but Prso insists he will be fit to play at Chelsea.
"I am keen to take some risk," said Prso.
"But there is not really any risk.
"For the moment I am doubtful, and despite all my desires, perhaps I will not be able to play.
"But I think it will be okay."
