The striker was making his comeback from a toe injury with Everton's reserves on Thursday evening.
However, just 22 minutes in he limped off with a calf injury - after he had earlier opened the scoring.
The 27-year-old will now undergo tests to gauge the full extent of the problem, although Everton are hopeful that the problem is not serious.
"James felt his calf tighten up after 15 minutes and he tried to run it off," Everton physio Mick Rathbone told the club's website.
"But he was still feeling some discomfort so he was sensible enough to come off the pitch.
"It looks like a relatively minor calf strain though it is too early to say whether he will be available for Spurs."
In better news for Everton both Per Kroldrup and Li Tie inch closer to full fitness.
Summer signing Kroldrup, who cost £5 million when he arrived from Udinese, is yet to make his Premiership debut after damaging his groin in pre-season.
He played in the same game as Beattie, but came through unscathed and could be in line for his first start against Tottenham next week.
Another candidate for the Spurs game could be Chinese star Li Tie.
The Far East ace last played in January 2004 when he suffered a cracked shin and his recovery has been slow, with a number of setbacks along the way.
Now Li Tie is hoping he can soon get his chance to stake his claim for a first team place.
"The manager told me I still need a little bit more practice.
"He doesn't want me to lose my confidence and he also said my chance may come any time.
"I am happy he said so. I know I have to be patient, but I also believe my opportunity will finally come.
"You have to realistic. I need more games to boost my overall fitness and skill, particularly making sure my touch returns 100 percent.
"I know what I have to do. I am just desperate to have a chance to play; as that is the only way I can improve myself.
"When you are in this situation you must be positive and get yourself ready all the time, because you never know when an opportunity will come around."
