Canaries boss Nigel Worthington has been chasing Hughes for much of the summer and has already had one bid rejected.
Reading had been hoping to persuade Hughes to sign an extension beyond his current deal, which expires in 2006, but they have now accepted Norwich's offer.
The figure has not been disclosed but The Canaries are now ready to start discussing personal terms with the 27-year-old.
Norwich's revelation that an accord has been reached with Reading could spark Championship rivals Burnley and Sheffield United into action as that duo are also keen on Hughes.
Meanwhile, Worthington is using the club's pre-season tour of Holland to cast his eye over PSV stopper Colin.
The 24-year-old has no future at Philips Stadion and has been training with the reserves while the first team are in South Korea.
He spent last season on loan at NAC Breda and will hope to convince Worthington that he merits a deal with Norwich.
