The 20-year-old has agreed a season-long loan deal at Layer Road.
U's have pounced for Vine to bolster their strike-force - which has already seen the likes of Craig Fagan and Wayne Andrews added to it this summer.
Vine spent most of last season on loan at Brentford and ended the season as the Bees' top-scorer, so he has already proved he is capable of finding the net in Division Two.
Manager Phil Parkinson was delighted to have landed Vine, saying: "There has been a whole host of Second Division clubs looking at Rowan during the close season.
"I rang Harry Redknapp way back in the summer about him and he said they were prepared to let him go somewhere for a year.
"However, they told me that I was going to face stiff competition because Brentford want him back and six or seven others wanted him.
"I spoke to the lad on a few occasions and I am really glad that he has decided to come.
"I think he has looked at the signings we have made in the summer, looked at things like the new stadium on Ceefax and TV and maybe he is thinking Colchester are a club who are going places."
