The 27-year-old initially joined the South West London club for pre-season training at the start of July and after two weeks with The Bees, manager Martin Allen has signed the Jamaican international to a one-year contract.
Burton arrives at the club on a free transfer after falling out of favour at Pompey last season and Allen was delighted to secure the services of a quality goalscorer like Burton.
"I am delighted we have been able to attract a striker of his calibre to Brentford," Allen told the Evening Standard.
"In the two weeks he has been on trial, he has done extremely well and he is joining as this club's number one striker.
"I am putting my faith in two forwards, Deon Burton and Isaiah Rankin, who can use Brentford as a springboard to climb back up the league."
Burton's signing comes after the club signed South Korean youngster San Lee from London rivals West Ham United this week.
