The two clubs had been locked in negotiations for several weeks regarding the possibility of Doherty moving to Ninian Park.
But the £1.3 million deal has failed to have been pushed through and Doherty will now stay at White Hart Lane.
The 23-year-old, who can play in defence or attack, has struggled to secure regular first-team opportunities in North London.
Doherty has featured just once for Spurs this term and a switch to South Wales would have kick-started his career.
Cardiff initially pulled the plug on their swoop for Doherty at the end of August but revived their bid to sign the ex-Luton ace until the latest setback.
The Irishman will now hope to impress Spurs manager Glenn Hoddle after being included in the squad for Saturday's clash against capital rivals Chelsea.
