A first half brace from James Beattie and then Kevin Phillips was enough for The Saints although Freddie Kanoute did grab a consolation.
The White Hart Lane faithful were silenced as early as the third minute.
Kasey Keller gave away a needless corner when he sliced a clearance and that error was punished when Matt Oakley's cross was met in fine style by Beattie.
The England star then showed his true class just before the interval when he stepped up and curled home a wonderful 30-yard free kick past Keller.
Tottenham introduced Robbie Keane and Darren Anderton at the interval and they added some impetus, but it was Southampton who extended their lead.
Phillips had been denied by the crossbar just before the hour, but he was on the scoresheet not long after as Chris Marsden and Graeme Le Saux worked the ball into the area and Anthony Gardner's clearance cannoned off the former Sunderland star and into the net.
The lively Kanoute grabbed his goal when he finished off a fine move involving Anderton and Keane, but it was too little too late for Spurs.
