Second half goals from Craig Beattie and Henri Camara settled the friendly, with Defendi appearing after the break for Jacques Santini's side.
Celtic looked sharper throughout, displaying the benefit of their opening league match at the weekend, as Kasey Keller saved from Camara early on.
French trialist Charles-Edouard Coridon was looking to impress Bhoys boss Martin O'Neill and showed flashes of ability in the game.
The opening goal arrived on the hour mark when the impressive Beattie, who was formerly a Spurs target, broke the deadlock.
Beattie picked the ball up and left the Spurs defence dazed and confused before slipping a shot past Keller.
Mark Yeates shaved the post for the Premiership side before Camara seized on a mistake from Goran Bunjevcevic to make it 2-0.
Bunjevcevic hit a weak backpass to Keller and Camara stole in to continue his fine start to his Bhoys career.
Camara almost made it three later in the game with a stunning effort from Beattie's cross which Keller pushed on to the post.
