The caretaker boss boosted his credentials by overseeing a thrilling victory at St Mary's, attracting the plaudits for utilising two strikers, something rarely seen under the charge of Andre Villas-Boas.
Holland manager Louis van Gaal is considered at 8/1 having revealed his desire to manage in the Premier League, although it looks unlikely that he would take the position before the World Cup given his reluctance to combine jobs.
Frank de Boer had headed the market but has now drifted out to 14/1 after reportedly ruling out leaving Ajax, while Fabio Capello follows at 20/1 having also been a favourite.
Sky Bet's Head of Football Trading Paul Wiggins said: "We already had Sherwood as marginal favourite for the job but Sunday's result and, possibly more importantly, the nature of it has seen the majority of the backing come for him.
"Given we settle our markets on the basis of being in charge for 10 games when there's no long-term appointment made, Sherwood looks the most likely pick given Spurs' willingness to give him a chance over a busy festive period."
