The England international was forced off at half time of Saturday's 2-0 InterToto Cup second leg win over Slovakian club ZTS Dubnica at St James' Park, as The Magpies cruised to a 5-1 aggregate success.
Jenas missed the first leg in Slovakia because of injury and Souness has confirmed he may miss the semi-final, first leg of their next InterToto Cup tie against Spanish giants Deportivo La Coruna.
"Jermaine is feeling a groin or hamstring but the physio feels it might be something which is more to do with his back," said Souness.
"At this point of the season, you can't afford to be pushing people at this stage.
"They have only been back a few weeks and you are risking a great deal asking him to play on in that situation."
Jenas's second half replacement in Michael Chopra also faces medical attention after being forced off himself with concussion against Dubnica.
"He thought he could continue, but he didn't really know where he was," Souness added.
"We will have to see what the doctors are telling us, but if you are concussed, you can't really rush back after that."
Despite the injury setbacks, Souness was pleased to see his side ease their way into the next stage of the summer European competition.
"We would have liked to have scored earlier today and made them sit back a bit more, but we were patient and got our rewards," Souness said.
"We have got to look at the big picture and we have won 5-1 in a tie after having been back in training for just two weeks."
