Tomlin initially joined Boro on loan from the League One side at the end of January but has now turned that into a permanent move.
The 25-year-old scored 10 goals for Posh this season and a total of 43 since his move from Rushden in 2010.
He is yet to make his Boro debut but could do so against Leeds United in the Sky Bet Championship on Saturday after completing a three-match ban.
Tomlin was sent off for the third time this season in Peterborough's 5-4 defeat by Oldham on January 25 and has not played since.
Watch Middlesbrough take on Leeds United on Saturday lunchtime, live on Sky Sports 1HD from 12pm.
