The Blades are preparing for the loss of Andy Gray to Sunderland and have turned to former Sheffield Wednesday striker Cresswell as a replacement.
But their efforts to tempt Preston into selling the 27-year-old have been swiftly rebuffed by their Championship rivals.
"We have turned down a substantial bid for Richard Cresswell from Sheffield United," Lilywhites chairman Derek Shaw told the club's official website.
"Richard has been our leading scorer for three of the last four seasons and he is not for sale."
Cresswell has formed an effective partnership with David Nugent in attack and manager Billy Davies is anxious not to lose any of his key players.
Davies also confirmed the bid from The Blades and revealed there has been an enquiry for Dickson Etuhu, with the midfielder having been linked with Everton.
The Preston boss said: "We have received an offer that we have turned down and we have also had an enquiry for Dickson Etuhu but that is as far as it goes."
