Michael Jackson was handed a second yellow card, after being adjudged to have fouled Adi Akinbiyi in the box - allowing John Eustace to smash home the penalty and cancel out David Healy's opener.
The game had already become a pitched battle as Stoke sought out a leveller, and the penalty left Preston's men fuming.
It had all looked so much better for North End when Healy had scored a fabulous goal for the Lancashire side.
The Northern Ireland man had lashed home from long range, after both Eddie Lewis and Ricardo Fuller had gone close.
Jonathan Gould had been in fine form throughout for Preston and denied Eustace on more than one occasion - notably in the closing stages.
Preston will rue the chance to put themselves back amongst the Division One challengers, whilst Stoke are a point further from the relegation quagmire.
