The Irish international strained his medial knee ligament against Preston on Wednesday and now faces four-to-six weeks out.
"Mark's received initial treatment and it appears he's strained the medial ligament in his knee," Wolves physio Barry Holmes told the club's website.
"We'll have the injury confirmed by scans and Mark will then see a consultant in Manchester on Friday and it's only then we'll know how long he'll be out for.
"But in our experience, such an injury usually takes between four and six weeks."
Defender Joachim Bjorklund also picked up a hamstring injury against Preston, so manager Dave Jones has promoted defender Keith Lowe and midfielder Kevin O'Connor into the first team squad from the club's youth ranks.
