Oakley sustained the injury during the dying stages of Saturday's loss to Boro and following a scan on the injury, he will be out for a lengthy period of time.
The news will come as a blow to Saints boss Gordon Strachan, with the in-form Oakley having been a key component of the South Coast club's positive start to the new Premiership season.
"It looked an innocuous fall but he has hurt the inside of a knee and will be a while," Strachan told the club's official website.
"It is very disappointing because he and Fabrice Fernandes have easily been our best midfielders this year."
Strachan will also be without the services of Graeme Le Saux for Saturday's trip to Newcastle United after he suffered a hamstring strain in training.
An ultrasound scan found his hamstring problem not to be healthy enough for this weekend and Le Saux will spend a week on the sidelines.
The defender will have a check on his fitness late next week in advance of the club's Uefa Cup first round, second leg trip to Steaua Bucharest.
"It is very frustrating. It just happened in training where I played the ball and turned and I just felt it go," revealed Le Saux.
"I am hoping I will be okay for the Steaua game but I will just have to wait and see.
"You never quite know with muscle injuries but it certainly won't be right for Saturday.
"The blank weekend for internationals will help. That will give me a bit of extra time to try and get fit."
