The 29-year-old former Chelsea stopper has inked a deal at the Britannia Stadium, having previously enjoyed a three-month loan spell there earlier in the season.
Duberry has endured a frustrating time at Elland Road since joining from Chelsea in 1999 for £4.5 million - failing to replicate the type of form that once made him a regular at Stamford Bridge.
After making just four appearances for Leeds this term, Duberry has decided to seek pastures new at Stoke.
In all competitions, Duberry made 76 appearances for Leeds - with injuries playing a part in his lack of participation.
Stoke director of football John Rudge has spoken of his delight at landing the dominating defender.
"Michael is set to sign for the club on a two and a half year deal,'' Rudge told the club's official website.
"We've come to an agreement with Leeds United and there will be no fee involved
"Michael had 18 months of his contract at Elland Road but he has reached an agreement to enable him to sign for us.''
