Bottom side Notts County earned a welcome first win of the season at Chesterfield while Swindon cut down high-flying Wrexham and Queens Park Rangers were held to a goalless draw at home to Wycombe.
First played third at Vale Park and it was the leaders at the start of the day who took the lead after just eight minutes as Rory Fallon put Barnsley 1-0 up.
Andreas Lipa levelled for Vale and goals from George Pilkington and Adrian Littlejohn gave the hosts a 3-1 win to send them to the top of the table and send The Tykes back down into third as they suffered their first loss of the campaign.
Sheffield Wednesday cruised into a 2-0 lead at Hillsborough against struggling Stockport County thanks to goals from Shefki Kuqi and Alan Quinn and looked to be sailing towards the top of the table.
However, Carlton Palmer was desperate to put up more of a show against his former club and his Hatters side came out after the break and earned a welcome point with two goals from Aaron Wilbraham securing a 2-2 draw.
Notts County are struggling both on and off the pitch this season but they received a welcome boost at Saltergate as Paul Bolland's strike gave them a 1-0 win against Chesterfield to hand them their first win of the season and also lift them off the foot of the table - to be replaced by The Spireites.
Wrexham's good recent run was halted at The County Ground as Swindon Town continue to impress by securing a 1-0 win courtesy of Andy Gurney's 79th-minute strike.
Richard Wellens gave Blackpool the lead at home to Bournemouth after 37 minutes but The Cherries came back in spirited fashion and goals from Stephen Purches and Karl Broadhurst gave them a 2-1 win at Bloomfield Road.
Plymouth also fell behind as Gary McSheffrey gave Luton Town the lead at Home Park but The Pilgrims also came back strongly and two goals from Micky Evans and David Friio gave them a 2-1 win.
Promotion favourites Bristol City took the lead at struggling Oldham through Tony Butler but Calvin Zola rescued a point for Iain Dowie's Latics.
Brighton came away from Layer Road with nothing as Wayne Andrews grabbed the only goal of the game to earn Colchester a 1-0 victory.
Stephen Hunt scored the only goal of the game at Nene Park as Brentford grabbed a 1-0 win against Rushden & Diamonds.
Last season's play-off finalists QPR were held to a goalless draw at home by Wycombe Wanderers while Tranmere were also held to a 0-0 draw at home by Peterborough.
