The 26-year-old was hoping to make a first team return with Phil Parkinson's side after six months on the sidelines with the knee injury.
Despite some recent setbacks, Duguid looked set for a return after taking an injection in his joint, but the effects of the jab have not helped improve his knee.
A specialist has told the former Layer Road trainee he needs surgery and another three months to recover.
"Doogie will have an exploratory operation on Monday to discover what is going on," U's physio Stuart Ayles told the club's official website.
"The surgeon will look inside and out of the knee as, so far, no one has been able to pinpoint the cause of the problem.
"The intention of the injection was to settle down the scar tissue and irritation that was lingering inside. It settled down initially, but as soon as we got back on the training ground and began kicking again, the problem flared up again."
