Peacock will be absent for at least two months with an ankle complaint and he is joined on the treatment table by loan signing Hasney Aljofree and Guy Branston.
Aljofree was new Owls boss Paul Sturrock's first signing when he was drafted in on a month's loan from the Scot's former club Plymouth Argyle.
But the defender has damaged his shoulder and is unlikely to feature again during his current spell with the South Yorkshire club.
Fellow stopper Branston has injured his calf and Sturrock is cursing the misfortune which has hampered his squad.
"The boy Peacock needs an operation and will be out for eight to ten weeks," Sturrock told the club's official website.
"Hasney has arguably been our best player in his three games. He has a tendon problem, it's detached off the bone in his shoulder.
"He'll be out for four to six weeks.
"We've found Guy Branston has a tear in his calf and there's no timescale on that."
