Argyle got off to the perfect start when Mickey Evans put them in front inside five minutes and Shrewsbury were reduced to ten men when Greg Rioch saw red for a second yellow card minutes later.
It was The Shrews' tenth red card of the season, but it did not affect them as Luke Rodgers (44, pictured), Sam Aiston (50) and Ryan Lowe (84) wrapped up the points and secured a league double against Paul Sturrock's side.
Luton missed the chance to take full advantage on Argyle's slip up though, as in-form Carlisle ended a fine week by earning a share of the spoils in a 1-1 draw at Kenilworth Road.
Steve Soley put The Cumbrians in front, but Russell Perrett earned Carlisle a point with a 67th minute equaliser.
Mansfield stay in third place after they eased past Hartlepool 3-0.
Graham Lee put through his own goal after three minutes and Wayne Corden wrapped up the points with the second 11 minutes from time.
Chris Greenacre then netted his 22nd goal of the season to cap a good day's work for The Stags.
Rochdale were held to a surprise 1-1 draw at home to Oxford despite taking an early lead through Patrick McCourt.
However Paul Moody grabbed a second half equaliser to earn a point for Ian Atkins' ever-improving side.
Pre-season promotion favourites Hull got their challenge back on track with a 2-0 win against Exeter and end The Grecians' stunning five-match winning run away from St James Park.
Julian Johnsson netted with a stunning volley before former Grecians hero Gary Alexander netted with his 20th goal of the season.
Steve Torpey's first half strike was enough for Scunthorpe in their clash with fellow promotion hopefuls Kidderminster, while Cheltenham remain amongst the play-off chasing pack with a 2-0 win at Leyton Orient.
Jamie Victory set them on their way with the first before Julian Alsop wrapped up the points for Steve Cotterill's side.
Rushden & Diamonds eased past midweek FA Cup heroes York City with a comfortable 3-0 win at Nene Park.
Duane Darby netted the first after three minutes and Michael McElhatton doubled their lead on the stroke of half time.
Onandi Lowe grabbed the third to cap a miserable day for The Minstermen, who had Chris Smith sent off for a professional foul.
At the bottom, Halifax picked up three much needed points with an impressive 3-0 win against Lincoln at The Shay to draw level on points with York.
The Shaymen have a good record against The Imps and they claimed the points through Gary Jones' double and Ian Fitzpatrick's late strike.
Darlington let a two-goal lead slip for the second time in a week as Southend fought back to claim a point in a 2-2 draw.
Paul Heckingbottom and Mark Ford (penalty) put The Quakers 2-0 up, but The Shrimpers hit back through Kevin Maher and Barrington Belgrave.
Chris Byrne's strike was enough for Macclesfield to claim a 1-0 win at Swansea, while Chris Brandon and Richard Logan scored the goals for Torquay as they came out on top in the West Country derby with Bristol Rovers. Nathan Ellington replied for The Pirates, who lost 2-1.
