The Tigers are now a point clear at the summit after the crucial victory at The KC Stadium.
After a goalless and tight first half, Hull went ahead just before the hour when Ben Burgess (pictured) scored and then a quick-fire brace from Australian Danny Allsopp (63 & 66) made sure of all three points.
Steve Basham pulled one back for Oxford with 13 minutes left, but then almost immediately Andy Crosby put through his own net, while Matthew Bound pounced in the last minute to give the score some respectability.
Doncaster took advantage of Oxford's defeat with a 2-0 win at lowly Southend United.
Both goals came in the opening ten minutes with Leo Fortune-West (4) and Michael McIndoe (7) giving them victory.
Mansfield Town also lost ground on the top two, after they went down 3-1 at Leyton Orient.
Gary Alexander put The O's ahead after just six minutes, but Jake Buxton made sure the scores were level at the interval.
After the break Orient took charge and goals from Tom Newey (70) and Alexander (82) made sure of victory.
Yeovil Town were the latest club to see at first hand Carlisle United's revival as they went down 2-0 at Brunton Park.
Player-boss Paul Simpson put his side in front after 21 minutes and former Leeds striker Craig Farrell made sure of victory 15 minutes from time.
The Cumbrians are now seven points off safety and just four points adrift of second bottom Darlington.
It could have been worse for The Quakers but they battled hard to secure a 1-1 draw at Kidderminster.
Adam Murray had put The Harriers ahead after seven minutes, but David McGurk pounced in the last minute to give David Hodgson's side a welcome share of the spoils.
Torquay United are right back in the play-off hunt after a good 3-1 win at Lincoln City.
Jason Fowler put The Gulls ahead after ten minutes, but Paul Mayo levelled just before the interval.
However, second half efforts from Kevin Hill (56) and Jo Kuffour (89) gave Torquay victory.
Huddersfield moved into the play-offs with a 1-0 win over Boston United at The McAlpine Stadium. Efe Sodje's goal with 19 minutes from time was enough to take the win.
Bristol Rovers came from two goals down to secure a 2-2 draw at Rochdale.
Patrick McCourt (18) and Kevin Townson (37) put their side two goals up, but a Matt Doughty own goal (74) and a Junior Agogo strike just seven minutes from time made sure it was all square.
Cambridge United eased away from the lower echelons of the table with a 1-0 win at Macclesfield Town. Highly-rated Luke Guttridge grabbed the only goal of the game after 49 minutes.
York City's promotion hopes were dented with a 2-1 defeat at Northampton.
Richard Walker put The Cobblers ahead after 20 minutes and their lead was doubled on half time when Jon Parkin put through his own net.
Lee Bullock pulled one back on the stroke of full time, but it was a mere consolation.
Scunthorpe against Bury and Swansea at home to Cheltenham ended goalless.
