The Tigers fully deserved their victory but had to work hard due to the fact that skipper Ian Ashbee was sent off just after the hour.
Despite being a man down Hull grabbed all three points thanks to a goal from Stuart Green on 64 minutes.
The game hinged around four second half minutes, which started in the 62nd minute as Ashbee produced a shocking challenge on Chesterfield's Adam Smith.
Referee Mark Cowburn wasted little time in producing a straight red card for the home skipper and sending him for an early bath.
Although Peter Taylor's men were a man light, they still managed to take the lead just two minutes after Ashbee's dismissal.
A good passing move saw Junior Lewis loft a ball into Green's path and he controlled but finishing well past Spireites keeper Carl Muggleton.
Taylor's men had to hang on in the dying minutes but they stood firm to register the three points to send them into second place.
Blackpool's recent improvement continued as they secured a 2-1 away win at Oldham Athletic.
The Tangerines made it two away wins on the bounce with success at Boundary Park, despite going a goal behind when William Haining struck on 39 minutes.
John Murphy levelled for the visitors two minutes before half time and Richard Wellens grabbed the winner on 68.
