The midfielder has signed a three-year contract with The Shrimpers after they agreed to pay an undisclosed fee to the League Two bottom club.
Guttridge had spent over four years at The Abbey Stadium and made over 150 appearances for the club.
The 22-year-old was taken aback by how quickly the move was agreed but had no hesitation in accepting a switch to Roots Hall.
"It came out of the blue to start with," Guttridge told Southend's official website.
"I've been trying to get out of Cambridge for a little while. When this chance came up I came down to the ground and met the lads, Steve (Tilson) and Paul (Brush) and I knew straight away that I wanted to sign.
"I decided on Southend because I was impressed with the management team and how they handled things. I like the atmosphere here already and the togetherness of the players."
Guttridge is due to make his debut for Southend in Saturday's League Two clash at Cheltenham Town.
