Luton boss Joe Kinnear was sent off after complaining bitterly that Mark Robins was offside when he ran onto a ball down the left and crossed for Sedgewick to finish easily.
However, Luton had plenty of effort in their attempts to equalise but could not way through a well organised Rotherham defence which moves them back into the automatic promotion place.
The Millers are now two points ahead of Reading and seem set for the first division for the first time since 1983.
Elsewhere, Craig Falconbridge's fourth minute strike saw the honours in the Welsh derby go to Wrexham at the expense of Swansea City.
Again there was a suspicion of offside as Kevin Cooper raced down the right and crossed for Falconbridge to side-foot home the only goal of the game.
Peterborough beat Walsall in the final game of the evening.
Jason Lee lashed in the first on the stroke of half-time when the visiting defence failed to clear a corner and Simon Rea's 57th minute effort secured the points for The Posh.
The game between Bristol Rovers and Port Vale was postponed because of a waterlogged pitch.
