The Welsh striker is yet to seal his City switch, although the newly-promoted outfit have been given permission for him to feature in the friendly.
Robins boss Gary Johnson has confirmed the 30-year-old, who is expected to seal his transfer on Monday, will play against Yeovil.
Johnson revealed to City's official website: "Lee will play on Saturday."
Swansea chairman Huw Jenkins could not contain his disappointment at losing Trundle.
"It's a very sad day for the club because we tried everything possible to keep Lee," he told Swansea's official site.
"We spoke about a wage increase amongst other incentives, but the one thing we couldn't guarantee him this season was Championship football."
