Lester completed his switch back to Forest from Sheffield United at the end of last month, and became an instant hero with the winner against Queens Park Rangers.
But he was stretchered off early in the second half of his third appearance for the club against Derby County on Saturday.
Scans have shown the 29-year-old has damaged his cruciate ligament and will miss the club's battle to stay in The Championship.
Under-fire Forest boss Joe Kinnear cursed the timing of the injury, particularly after Lester's fine start to the second chapter in his City Ground career.
"It's a terrible blow, most of all for the lad but for the club as well," Kinnear told the club's official website.
"He had settled straight back in after returning to The City Ground from Sheffield United and made the kind of impact we hoped he would. Then this happens.
"He will be back but we just have to get on with life without him for the rest of the season."
