The Swans included six loan players in their 16-man line-up during last Saturday's Welsh derby against Wrexham at Vetch Field.
Neil Cutler, Leon Hylton, Alan Tate, Leon Britton and Marc Richards were all used by Swansea director of football Brian Flynn on Saturday - while the sixth loanee Craig Stiens was an unused substitute.
The Football League introduced new regulations at the beginning of the season, which state that a club can only name five loan players in their squad for any given match.
Under the new regulations, Swansea now face some form of punishment, with a fine or even points in the table being deducted for their indiscretion.
"We are asking the club for an explanation," said a Football League spokesman.
"There is a possibility that they'll be some sort of hearing into this because it is a breach of regulation.
"Having said that, it's not a major breach of regulation, mainly because the sixth player didn't actually take the field of play."
Meanwhile, The Swans have extended the loan of defender Leon Hylton by another month from Premiership side Aston Villa.
