The 38-year-old was already considered the frontrunner to replace Guy Whittingham before leaving the Reds but has had his price cut further with Sky Bet following persistent backing.
Barker was relieved of his duties at Crawley following a seven-match winless run, in which his side scored just one goal and slipped down to 12th in Sky Bet League 1.
The former Rotherham striker is followed in the next Portsmouth manager betting by Martin Allen (5/1), Neil Warnock, Mikey Harris (both 7/1), David Connolly, Justin Edinburgh and Chris Wilder (all 9/1).
Sky Bet Football Trader Chris Spicer said: "It was somewhat of a surprise to see Barker backed into favourite for the Portsmouth vacancy given he was already in a job and certainly less of one to see him part company with his previous employers.
"Whether that money came on the back of suggestions that his position was under threat or genuine links with Pompey remains to be seen, but the one-sided backing has forced us to keep our price short, with Barker attracting over half of the money in our book."
Meanwhile, striker Gary Alexander and chief scout Martin Hinshelwood have been placed in interim charge at Crawley, with the former currently 2/1 favourite to take the position on a full-time basis.
