Hearts looked set for an important win after racing into a 2-0 half time lead, but Celtic fought back in the second half to claim an unlikely victory with McManus the hero with two late strikes.
Stilian Petrov wasted a golden opportunity to give Celtic the lead after just four minutes when he headed Alan Thompson's cross over the bar from just six yards out.
Hearts made Petrov pay for that miss two minutes later when Paul Hartley broke down the left and his dangerous cross was turned into his own net by the unlucky Ross Wallace.
The home side were in dreamland on eight minutes when Steven Pressley headed home Rudi Skacel's corner.
Celtic tried to fight back and Shaun Maloney and Petrov both saw long-range strikes saved by Craig Gordon.
Hearts missed a great chance to go 3-0 up a minute before the break when Skacel saw his point-blank shot well kept out by Artur Boruc.
Celtic suffered a blow minutes after the break when Petrov was forced off with a hamstring complaint.
His replacement Stephen Pearson pulled a goal back for Celtic on 55 minutes when he slotted home from Maloney's cross.
Hearts were reduced to ten men on 76 minutes when Takis Fyssas was sent off for hauling down Maloney as he raced through on goal.
McManus drew Celtic level three minutes before the end when he headed home Shunsuke Nakamura's inviting free kick.
The big defender then grabbed the winner deep into injury time when the ball fell nicely to him inside the box and he turned and fired a shot past Gordon to send the visiting fans into raptures.
