Lyon needed victory to move above Monaco and they gained all three points at Stade Gerland.
Strasbroug had looked as they would hold on for an admirable point, but two minutes into stoppage time defender Fabrice Ehret handled the ball in the area.
Ehret received a second yellow card and was dismissed and then to pile on the misery, Luyindula stepped up and beat Richard Dutruel from the spot.
Lyon had earlier been reduced to ten men when Mickael Essien was dismissed for his second yellow card on the hour, but the home side pushed forward and gained their winner - albeit with just seconds remaining.
l'OL are now level with Monaco at the top, but ahead on goal difference - however the Principality outfit will move back top if they avoid defeat at home to Lens on Saturday.
