The high-flying Primera side were given the perfect start by Carlos Aranda who fired them ahead after just seven minutes.
It was a lead they never looked like surrendering, and it was something of a surprise they did not extend their advantage until 13 minutes from time.
Their second came courtesy of prolific Brazilian Julio Baptista (pictured) who coolly converted a penalty.
Lille made up for defeat in their opening game by edging past Zenit St Petersburg.
All three goals of a tight encounter arrived in a seven minute spell towards the end of the first half.
Gregory Tafforeau moved the hosts ahead on 35 minutes, but Alexander Kerzhakov responded just four minutes later.
However, the game was settled three minutes before the break when Matt Moussilou grabbed the goal which reinvigorates Lille's hopes of progression.
