The Russian international was not the only hero for Celta as keeper Pablo Cavallero saved Rivaldo's injury-time penalty to give the home side a good chance to get into the final.
Barcelona, however, had stormed into an early lead, Simao Sabrosa tapping home Boudewijn Zenden's pass after just six minutes.
This stung Celta into action and they launched wave after wave of attack on the Barca defence and eventually breached it a couple of minutes from the interval.
Eduardo Berizzo smashed in a superb effort from Mostovoi's free-kick to send the sides into the break all square.
Mostovoi then put his team into the lead in the 51st minute when Barca's defensive frailties were exposed and the midfielder was free to glance in a superb header from Juan Velasco's cross.
With 18 minutes to go a great one-two between Mostovoi and Valery Karpin set-up Jesuli who beat Manuel Reina with a brilliant effort.
The final ten minutes were a tense affair for the home side as both Fernando Caceres (82) and Juan Velasco (88) were dismissed and Rivaldo missed his spot-kick and Celta clung on for the win.
Barcelona now face a tough task on Sunday to overhaul the deficit and qualify for the final.
