The top of the table clash was keenly anticipated in Spain and it was Barca who took the lead as Ronaldinho converted from the penalty spot 28 minutes in.
The lead lasted less than ten minutes, though, as former Tottenham and West Ham star Fredi Kanoute levelled.
Ronaldinho then restored Barca's lead just before the interval and the win was sealed late on with Argentine starlet Lionel Messi firing home.
The win means Barca are now three poinst clear at the top, after Valencia lost 3-2 to Celta Vigo - their first loss of the season.
Los Che took the lead at The Estadio Balaidos through David Navarro, but Celta levelled just before the break through Fernando Baiano.
Celta found themselves two goals to the good as they fired home twice in four minutes through Iriney (52) and Nestor Cannobio (56).
Fernando Morientes gave Valencia some hope with his strike on the hour, but they could not find a way through and their misery was compounded in the final minute when Jaime Gavilan was dismissed.
Real Sociedad remain rooted at the bottom of the table after losing 3-1 at home to Real Zaragoza.
Sociedad's day got off to a poor start when Diego Rivas was sent off for two yellow cards on the half hour. Then minutes later they were behind as Diego Milito converted from the penalty spot.
Xabier Prieto's penalty (42) gave the home side some hope, but second half efforts from Carlos Diogo (55) and Milito (59) gave Zaragoza the victory.
Sociedad are now the only win-less side in Spain after both Atheltic Bilbao and Racing Santander secured victories.
Bilbao grabbed a 3-2 victory at Gimnastic De Tarragona, while Racing beat Osasuna 1-0 in Navarra.
Francisco Yeste put Bilbao ahead after just 11 minutes and their hopes of victory were enhanced when Manuel Ruiz saw red for Gimnastic.
Andoni Iraola (29) added another before the interval, and then Yeste converted again on 50 minutes.
Josu Sarriegi (55) put through his own net and Ariza Makukula's (71) strike gave the home side real hope but they could not find a leveller.
Racing win came courtesy of the penalty spot as Felipe Melo converted just after the interval.
Real Mallorca sealed a 1-0 victory at Levante thanks to Juan Arango (27). The home side had Olivier Kapo dismissed 14 minutes from time.
Villarreal remain way off the pace after being held to a goalless draw by Espanyol.
The final game of the day saw Real Betis claim a 1-1 draw with Deportivo La Coruna thanks to a last-minute penalty.
Betis had looked to be heading for defeat after a Javier Arizmendi strike just moments into the second half.
Then in injury time, after both sides had been reduced to ten-men with Rafael Sobis and Luis Kasmirski seeing red, Betis were awarded a penalty when Albert Lopo handled a cross, and Robert de Pinho de Souza took the chance to level from the spot.
