The Argentine ace's current deal was due to expire in the summer, but the club have moved to tie him down to a new contract.
The 32-year-old, who has formed a formidable central defensive partnership with Roberto Ayala, was delighted to have sorted out his future.
"I have agreed to stay for another year," said Pellegrino.
"I'm very happy that both the club and [coach Rafael] Benitez were keen to see me continue here."
Meanwhile, Valencia have suffered a blow with the news that reserve goalkeeper Andres Palop will miss the rest of the season through injury.
The 30-year-old sustained a hand injury while warming up for last week's Uefa Cup tie with Genclerbirligi.
Tests have shown he needs to undergo surgery after damaging ligaments which will sideline him for up to two months.
Palop's injury leaves Valencia with only David Rangel as cover for number one keeper Santiago Canizares.
