The Argentine coach rested two of his fellow countryman, adjudging Hernan Crespo too tired to feature at all, and naming Javier Zanetti as a substitute.
The decision has already been criticised, but Cuper insisted that he was not prepared to risk tired players in the game - ahead of the Champions League clash with Newcastle United, although he did play another Argentinian international Nelson Vivas from the start.
"It is my fault - that is all I can say," said Cuper after watching a goal from Serginho sink his side.
"When you lose a match a coach has to take responsibility - so I was wrong.
"It's easy making decisions in retrospect. I had to make the choices before the game.
"There were difficult decisions. Dropping Crespo for instance.
"He was very tired and I thought that it would not be clever to risk a forward in this condition.
"Zanetti wasn't fit enough to play a whole match so I decided to leave him until the second half.
"I know the derby is a fundamental match, but we also have to look to another fundamental match in a short time - against Newcastle United."
