Marcelo Lippi's side ran out to a 2-0 half time lead as Fabrizio Miccoli scored after Paolo Montero played the ball to him from Stephan Appiah's corner before Gianluca Zambrotta hit a wonderful half-volley strike from Miccoli's corner two minutes before the break.
Three minutes after half time, Miccoli scored his second with a low header on his knees in front of Ancona's goal after Zambrotta sent in a fine cross from the left.
With Juve in a comfortable three-goal lead, the home side started to come back into the match and William Viali would make it 3-1 with a fine near-post header after substitute Maurizio Ganz played a cross in from a short corner.
Then, as the match went into injury time, Ganz scored Ancona's second with a fine left foot finish past Gianluigi Buffon to make the five minutes of injury time interesting but Juve were able to hold on for the three points.
In the earlier Serie A match on Saturday, Internazionale got a late Christian Vieri penalty with three minutes remaining to earn a 2-2 draw with Brescia at the Stadio Mario Rigamonti.
The home side, however, seemed certain to draw level on points with Inter in the table after Roberto Baggio gave Brescia a 1-0 half time lead and Andrea Caracciolo made it two three minutes after the break.
But substitute Julio Cruz pulled one back for Hector Cuper's men four minutes after coming on and Vieri would then equalise from the penalty spot to salvage an away point for the Milanese outfit.
