Fabio Capello's men cemented their place at the summit of the table after securing a 1-0 victory over Reggina.
Del Piero took his Juventus tally to 186 goals as he struck on 45 minutes to give The Bianconeri all three points at Stadio Delle Alpi.
The evening clash saw Milan fail to reclaim second place as they went down 1-0 at Roma.
Brazilian Alessandro Mancini grabbed the only goal in the Eternal City just nine minutes from time - the victory sees Roma move into sixth.
Milan's loss meant Internazionale stayed in second, after they had earlier sealed a thrilling 3-2 victory over Cagliari - who move into the relegation zone following the result.
The Nerazzurri opened up a two goal lead thanks to early strikes from strike partners Obafemi Martins (10) and Adriano (14), although Mauro Esposito halved the deficit on twenty minutes.
Inter restored their two-goal advantage when Brazilian star Adriano grabbed his second from the penalty spot (58), and the goal proved crucial as David Suazo netted with nine minutes to go.
Fiorentina also took advantage of Milan's loss as they moved into third thanks to a last gasp strike from Luca Toni gave them a 2-1 win over Chievo.
The Italian international took his Serie A tally for the season to 18 as he grabbed a brace, and the second of the two proved all-important after Andrea Zanchetta (71) had scored for the visitors from Verona.
Livorno dropped two points on their Florentine rivals but remain in fifth after a 2-2 draw with Siena.
The visitors took a shock lead at Armando Picchi through Tomas Locatelli (8), although Stefano Morrone (24) drew Livorno level.
The hosts edged ahead through star striker Cristiano Lucarelli (52), but Siena earned a point when Enrico Chiesa scored from the spot five minutes later.
Parma moved out of the relegation zone after a 1-1 home draw with Lazio.
Tommaso Rocchi had put the capital club ahead after just five minutes, although Bernardo Corradi punished his old side as he equalised on 38 minutes.
Udinese moved into the top half of the table after being held to a 2-2 draw by Treviso.
Last season's UEFA Champions League qualifiers took the lead through Marco Motta (13) before Pinga (33) and Pablo Dellafiore (58) handed the league's bottom side an unlikely advantage.
However Antonio Di Natale saved the hosts from embarrassment as he scored from the penalty spot on 72 minutes.
In Serie A's other clash Ascoli beat ten-man Empoli to leapfrog their opponents in the table.
The home side sealed the win thanks to goals from Marco Ferrante (37), Sasa Bjelanovic (70) and Maurizzio Domizzi (76), whilst Empoli netted through Sergio Almiron (45) before seeing Andrea Raggi dismissed.
