The Giallorossi skipper sustained a knee injury during a training session as the club prepared for their latest match on their USA tour against Liverpool.
Totti immediately underwent scans on the injury which revealed a knee ligament problem that could require surgery.
The Italian international has flown back to Rome for further tests to find out the full extent of the problem and decide whether or not he needs to go under the knife.
Should Totti require surgery he is expected to be out of action for up to a month, ruling him out of the start of Serie A in September.
Totti remains hopeful that the injury is not too serious and that he will be back in action soon.
"I'm upset. Anyway it seems it is not a serious thing and even if I need surgery I'll have an absence of about a month," said Totti.
Totti's problem is the second major injury blow for Roma this pre-season, as Damiano Tommasi faces up to a year on the sidelines after picking up a cruciate knee ligament injury during his side's friendly with Stoke City.
